When human peripheral blood mononuclear cells (hPBMCs) are exposed to certain ODNs, a cascade of immune mediators is unleashed, including interferon gamma (IFN-γ), tumor necrosis factor alpha (TNF-α), and granulocyte/monocyte colony-stimulating factor (GM-CSF). However, this symphony of cytokine release requires the presence of interleukin 2 (IL2), the maestro of immune activation.

GM-CSF: A Spotlight on CD56+ Cells
GM-CSF secretion, a key aspect of immune defense, was found to be particularly robust when non-CpG ODNs with high T content and PyNTTTTGT motifs were employed. Conversely, CpG ODNs and IFNα, another immune regulator, acted as antagonists, suppressing GM-CSF production.
Intriguingly, selective removal of cell types from hPBMCs revealed that CD56+ cells were the maestros behind GM-CSF secretion, while plasmacytoid dendritic cells (PDCs) played a regulatory role in this process.
A Tale of Two Oligonucleotides: PyNTTTTGT and CpG
PyNTTTTGT ODNs, in a surprising twist, were found to inhibit IFNα secretion induced by CpG ODNs in PDCs. This interference stemmed from their ability to disrupt the TLR9 signaling pathway, a crucial communication channel for immune activation.
Given that IFNα is essential for CD56+ cell stimulation by CpG ODNs, this reciprocal interference between PyNTTTTGT and CpG ODNs highlights the intricate interplay between these synthetic molecules and the immune system.
